Herpes and other viruses that attack the nervous system may thrive by disrupting cell function in order to hijack a neuron's internal transportation network and spread to other cells.
Princeton University researchers made the first observation in neurons that common strains of the herpes virus indirectly take control of a cell's mitochondria, the mobile organelles that regulate a cell's energy supply, communication with other cells, and self-destruction response to infection. The team reports in the journal Cell Host and Microbe that viral infection elevates neuron activity, as well as the cell's level of calcium a key chemical in cell communication and brings mitochondrial motion to a halt in the cell's axon, which connects to and allows communication with other neurons.
The authors propose that the viruses then commandeer the proteins that mitochondria typically use to move about the cell. The pathogens can then freely travel and reproduce within the infected neuron and more easily spread to uninfected cells. When the researchers made the mitochondria less sensitive to calcium the viruses could not spread as quickly or easily.
These findings reveal a previously unknown and highly efficient mechanism that some of the most common strains of herpes viruses in humans may use to proliferate in the nervous system, said lead author Tal Kramer, a doctoral student in the lab of the paper's co-author Lynn Enquist, the Henry L. Hillman Professor of Molecular Biology and chair of Princeton's molecular biology department.
Kramer and Enquist used rat neurons to study two herpes viruses in the alpha-herpes virus subfamily: pseudorabies virus (PRV), a model alpha-herpes virus that infects animals, and herpes simplex virus 1 (HSV-1), an extremely common human virus that causes cold sores and other lesions. Other human alpha-herpes viruses are responsible for causing diseases such as chicken pox and shingles.
"No one before has looked carefully at mitochondrial motion during alpha-herpes virus infection in neurons. We provide new insight into how these viruses damage cells in the nervous system in ways that are important for the virus to propagate," Kramer said.
"If mitochondria are stopped in their tracks and can't go anywhere, that is potentially very bad," he said. "They are not only the power plants of the cell, but regulate important processes. The virus likely acts to interfere with many of those processes."
Beyond herpes, the Princeton findings present a possible explanation for how other neurotropic viruses such as rabies, West Nile and polio attack and disrupt the nervous system, Kramer said. Although these viruses are different from the herpes family, the fact that HSV-1 and PRV had a similar effect on mitochondrial motion and function suggests that other pathogens could corrupt mitochondria in the same way, he said.
In addition, the paper lays out the implications of distorted mitochondrial function on neuron health. Mitochondrial malfunction is a known factor in non-infectious neurodegenerative conditions such as Alzheimer's disease and Parkinson's disease, Kramer said, though the pathway to this disruption is not entirely known.
"Our model raises some new and exciting possibilities for future research on other important human viruses that can invade the nervous system and cause disease," Kramer said.
"And the fact that alpha-herpes infection damages the same key cellular function as neurodegenerative disorders also is striking," he said. "Understanding how viral infection damages neurons might give us insight into how diseases like Alzheimer's do the same. The viruses we study hijack well-studied cellular pathways that might make an effective target for future therapeutic strategies."
In a healthy neuron, mitochondria move throughout the cell's elongated, tree-like structure to provide energy for various processes that occur throughout the cell. For the strenuous task of long distance intercellular communication, mitochondria move along the axon and synapses, sites of cell-to-cell contact where signaling occurs.
Calcium plays a key role in this cell communication, Kramer explained. A neuron experiences a spike in calcium levels in the axon and synapses when it receives a signal from another neuron. Though a natural rover, mitochondria contain a protein called Miro that detects this rush of calcium and stops the organelles in the synapse. The mitochondria then provide energy as the cell passes a signal along to the next neuron.
Through live-cell imaging of neurons grown in the Enquist lab, Kramer and Enquist observed how this process becomes corrupted by HSV-1 and PRV and how the viruses need the process to spread.
The chaos begins when the virus ramps up the neuron's firing of electrical signals, as was first reported in a 2009 paper published in the journal PLoS Pathogens by Enquist; first author Kelly McCarthy, a past member of Enquist's lab who received her doctoral degree from Princeton in 2011; and David Tank, the Henry L. Hillman Professor of Molecular Biology and co-director of the Princeton Neuroscience Institute.
In the latest research, Kramer and Enquist found that this spike in electrical activity floods the axon and synapses with calcium. As a consequence, the Miro proteins detect the increase in calcium and stop mitochondrial motion. The virus' control over the cell immediately dropped off, however, when Kramer and Enquist interfered with Miro's ability to respond to the uptick in calcium levels. Though the viral infection was not completely disrupted, it could not spread within or to other cells with the same efficiency.
Based on these observations, Kramer and Enquist suggest that viruses such as HSV-1 and PRV may bring mitochondria to a standstill in order to hijack their transportation. Mitochondria move about the neuron on the backs of motor proteins dynein and kinesin-1. During viral infection, mitochondria shed these proteins to stop moving when Miro detects an upsurge in cellular calcium.
Previous research has shown that HSV-1 and PRV also use kinesin-1 specifically for transport within an infected cell. Thus, Kramer said, his and Enquist's work suggests that it is very likely that the viruses disrupt mitochondrial motility so that they can hitch themselves to the now available kinesin-1 proteins and move through the nervous system more efficiently.
James Alwine, a University of Pennsylvania professor of cancer biology, said that the Princeton research is a significant contribution to a growing body of research that describes how viruses seize cellular motor proteins such as kinesin-1.
While the findings have therapeutic potential particularly in helping show how balancing cellular calcium might subdue viral infection the demonstration that viruses can move through an infected cell with the ease of something as essential as mitochondria is notable in itself, said Alwine, who is familiar with the research but had no role in it.
"Determining the specific mechanism by which Miro function is abrogated may provide additional therapeutic avenues, but this also is marvelous basic research that does not have to be justified by its therapeutic potential," he said.
"To disrupt the loading of mitochondria to motor proteins so that virions [complete virus particles] can load instead is a clever way for a virus to be transported and is a great new idea provoked by this data," Alwine said. "While other neurotropic viruses would have to be tested specifically, movement in nerve cells is required by all of them. Thus, this observation provides a starting place and a model mechanism for research with those other pathogens."

SEOUL, South Korea (AP) ? Samsung Electronics Co., the world's largest maker of mobile phones, said its third-generation Galaxy S smartphone went on sale Tuesday in 28 European and Middle Eastern countries, hoping to cement its lead over Apple's iPhone.
The Galaxy S III smartphones hit the shelves in countries including France and Britain. By the end of July, nearly 300 mobile operators around the world will be selling the device.
Samsung said the initial response to the new smartphone was positive. At Vodafone Group, pre-sale orders for the Galaxy S III outstripped those of any previous Android-based smartphones, according to Samsung.
The Galaxy series of smartphones is widely acknowledged in the industry as a success for Samsung, turning it from a smartphone also-ran into a viable competitor with the iPhone from Apple Inc.
While Apple has kept the screen size of the iPhone the same in every upgrade since 2007, Samsung increased the screen size of its highest-end smartphone in its two major updates. The latest Galaxy phone screen is nearly twice as big as the iPhone screen.
The 4.8-inch S III also features more computing power that supports voice commands and an eye-movement tracking feature to keep the screen from dimming.
Samsung overtook Apple in smartphone sales for the first time in the first three months of this year, according to research firm Strategy Analytics. The Suwon, South Korea-based company sold 44.5 million smartphones in the January-March quarter, compared with Apple's 35.1 million iPhone sales.
Market watchers expect that Apple may use its annual conference for developers next month to announce the yearly upgrade to the iPhone, which then can go on sale as early as July.
Aside from being big rivals in the smartphone market, Samsung and Apple have also a close business relationship. Samsung supplies mobile chips and display panels for iPhones and iPad tablet computers, counting Apple among its biggest clients.

I first encountered No Longer Empty in Howard Hurst's Hyperallergic article from early May. Intrigued by the seemingly impossible partnership between "pop-up" exhibition-making and community building, I sought out Executive Director Naomi Hersson-Ringskog to inquire further into the nonprofit, beyond what could be found on their website.
Given its nomadic nature, weaving in and out of neighborhoods and communities throughout New York City, the No Longer Empty team inhabits the space in which each project is produced, which is precisely the reason Ms. Hersson-Ringskog and I met at the Bronx's own Andrew Freedman Home -- a place for old folks who'd lost their fortunes to sustain their lavish lifestyles -- and the setting for NLE's current exhibition, This Side of Paradise.
Upon entering the grand Home, I was greeted warmly by curatorial volunteer, Lucy Lydon (a gesture apparently foreign to many galleries and museums here in New York). I floated through the bottom two galleries, primarily visiting with Mel Chin's S.O.S. Reloaded: Bronx piece -- video shots of Bronx residents, their personal messages to President Obama tickering across the bottom of the screen, while heartbeats compose a rhythmic soundtrack.
I quickly noticed synergies running through the exhibition. Each site-specific work speaks individually yet collectively to themes of nostalgia for a no longer existent past through representations of the present; histories and remembrances of the Home; the social, political, and cultural significance of the Bronx; and the illumination of a space within a community.
Soon after my time spent on the bottom floor of the show, Ms. Hersson-Ringskog and I claimed a section of grass on the front lawn of the mansion and discussed NLE's process and challenge of building professional exhibitions, while remaining grounded within community.
Each project begins with extensive, organic research, partnering with local community organizations, community boards, and business improvement districts, to name a few. "The exhibitions are a way of paying homage to the site and to the neighborhood," Hersson-Ringskog explained.
Not unlike other projects NLE has completed in the past, This Side of Paradise represents a mechanism for cultural tourism and encourages further exploration into local cultural centers and neighborhood businesses in the Bronx. At the information desk of the Andrew Freedman Home, you will find a brochure described as "a cultural map of places, people & curious histories." Inside is a detailed map of the Bronx, identifying destinations such as, Bronx Art Space, Longwood Arts Project/Gallery at Hostos Community College, and Bronx Music Heritage Center; while marking the "stomping grounds" of well known people, like Grandmaster Flash, Edgar Allen Poe, and Collin Powell.
Alongside their exhibitions, the NLE team collaborates with local groups and people to produce programs that speak to the themes of each exhibition and the environments in which they live. "It's about activating the site," Hersson-Ringskog said when I asked about the programs. While in partnership with The Bronx Arts Alliance and collaborating with groups including The POINT and the Bronx River Art Center, the list of programs for This Side of Paradise has grown organically throughout the course of the exhibition and helped establish what Hersson-Ringskog calls, "inroads with the community."
Perhaps the most impressive characteristic of this three-year old nonprofit is its pervasive consistency in regards to its mission and vision. From its free admission and open-door policy, to its desire to gather community participation and visitor response, No Longer Empty remains an innovative cultural organism in which creative practice, professional curation, and community vitality are fused and enriched within the diverse neighborhoods of New York City.
In this downtrodden economy in which the arts are often seen as superfluous and secondary, it is inspiring to see a young arts organization thriving and actively changing the ways in which people engage with nomadic art making and acknowledge the spaces and places in which art is created and shared.
On the subway ride downtown I shared a seat with Lisa Kahane, This Side of Paradise contributing artist and principle photographer of the one and only Fashion Moda. She was kind enough to share her experience in the exhibition. "Working with the team at No Longer Empty is easy," Kahane says. "The women I work with -- Manon, Naomi, Jodie, Charlotte and Lucy -- are unfailingly supportive and patient, even under duress, working overtime to make things happen. When new situations come up during the show, they incorporate them into the whole. Most importantly, everyone understands that the interplay between art and context increases meaning."
Having photographed the Bronx since 1979, Kahane has seen and documented the rapid changes the borough has endured over time. Kahane's living project for This Side of Paradise includes a "traveling portrait studio" and visual documentation of the space, people, and community engaging with the exhibition. You can find her in Room 248 in the old home, and depending on the circumstance she may even take your picture.
This Side of Paradise will be open to the public until June 6. Don't miss your chance to experience a visual labyrinth where original works of art speak to the histories, architecture, and people of a given space.

If there is any issue that threatens to derail the fragile East-West d?tente that's held since President Obama set out to reverse the mini-cold war that prevailed under George W. Bush, it's the increasingly acrimonious dispute over missile defense.
Earlier this month Russia's top general, Nikolai Makarov, went so far as to suggest that his forces might launch a preemptive strike against NATO missile defense emplacements in Central Europe?if they were perceived to threaten Russia's strategic nuclear deterrent.
But NATO, which has made the issue a litmus test of alliance unity, has remained unmoved by Russian bombast on the subject and is clearly moving forward with the project, which is planned to reach full operational capability by 2020.
"This is the first step towards our long-term goal of providing full coverage and protection for all NATO European populations, territory, and forces. Our system will link together missile defense assets from different Allies ? satellites, ships, radars, and interceptors ? under NATO command and control," NATO Secretary General Anders Fogh Rasmussen told the summit on Sunday. "It will allow us to defend against threats from outside the Euro-Atlantic area," he added.
Buried deep inside NATO's Chicago Summit Declaration is the strongest political statement yet offered by the alliance in hopes of mollifying Russian worries: "NATO missile defense is not directed against Russia and will not undermine Russia?s strategic deterrence capabilities," it says. "While regretting recurrent Russian statements on possible measures directed against NATO?s missile defense system, we welcome Russia's willingness to continue dialogue with the purpose of finding an agreement on the future framework for missile defense cooperation."
While that statement may be perceived in Moscow as progress, it falls far short of the legally binding written pledge that the Kremlin has demanded.
"We've heard this before," says Fyodor Lukyanov, editor of Russia in Global Affairs, a leading Moscow-based foreign-policy journal. "The thing is, when the?Americans say their missile defense plans are not directed against Russia, they're telling the truth. It's against everybody. Since Ronald Reagan first floated the idea, missile defense has been seen as a way to protect the US against any and every possible missile threat. But Russia is the main country whose national security is based on strategic nuclear deterrence, in a balance with US forces. It cannot help but concern us directly."?

It?s being called a national epidemic: obesity in the state of Alaska. And officials say it?s hurting our kids.
Three out of every ten Alaska kids are obese, and will more likely grow up to be obese adults dealing with major illnesses. Fighting obesity has become a top priority for health advocates who are using a powerful documentary to get families off their couch and moving.
With 30 percent of kids in Alaska considered overweight, it?s what could happen when they?re older that health officials say will cost us more than we already spend.
?Heart disease, stroke, diabetes, arthritis,? said Karol Fink, who is the Obesity Prevention manager for the State of Alaska. ?We?re spending $459 million annually on medical care costs attributed full to obesity just in Alaska.?

PROVO, Utah (AP) ? Former major leaguer Vance Law is out as BYU's baseball coach after 13 seasons.
Athletic director Tom Holmoe announced Friday that the school will not renew Law's contract for next season. The former infielder is 397-347-2 with the Cougars entering the team's season finale against Gonzaga on Saturday. BYU is 22-26 this season.
Holmoe said assistant coaches Ryan Roberts and Wally Ritchie will take over the program while the university searches for a replacement for Law.
Law played at BYU in the mid-1970s and spent time with Pittsburgh, the Chicago White Sox and Cubs, Montreal and Oakland from 1980-91, and also played a season in Japan. He made the NL All-Star team in 1988 with the Cubs.
